糖新生の律速酵素である phosphoenolpyruvate carboxykinase (PEPCK) のインスリンによる遺伝子発現調節機構を検討した。PEPCK遺伝子の転写調節領域とCATレポーター遺伝子とを融合させたプラスミドを安定的に導入した肝培養細胞であるHL1C細胞を dexamethazone および8CPT-cAMPで刺激するとPEPCK遺伝子転写活性は約50倍の増強をみたが, この増強はインスリン処理により約75-90%抑制された。アデノウイルスベクターを用いた dominant negative phosphatidylinositol (PI) 3-キナーゼの導入により, このインスリン効果はほぼ完全に消失した。Dominant negative Sos の導入により, インスリン依存性のMAPキナーゼの活性化は完全に抑制されたが, インスリンのPEPCK遺伝子転写に対する効果は影響を受けなかった。さらに, constitutive eactive PI 3-キナーゼを導入するとインスリン非存在下でもPEPCK遺伝子転写は抑制された。以上の結果より, インスリンのPEPCK遺伝子転写抑制効果にはPI 3-キナーゼの活性化が必要かつ充分であり, RASにより活性化される情報伝達経路はこれに関与しない可能性が示唆された。
Phosphoenolpyruvate carboxykinase (PEPCK) is a key enzyme that regulates gluconeogenesis in the liver. Insulin decreases PEPCK activity in the liver by inhibiting its transcription, subsequently results in the inhibition of gluconeogenesis. Regulatory mechanism of the inhibitory effect of insulin on PEPCK gene transcription was investigated with the use of adenovirus vectors encoding a dominant negative or a constitutively active mutants of phosphoinositide (PI) 3-kinase, or a dominant negative mutant of Sos, an activator for RAS. Insulin treatment resulted in the inhibition of PEPCK gene transcription promoted by 8CPT-CAMP and dexamethazone. Overexpression of a dominant negative mutant of PI 3-kinase almost completely abolished the insulin's effect on PEPCK gene transcription. A dominant negative mutant of Sos did not affect PEPCK gene transcription whereas the mutant Sos abolished insulin-induced MAP kinase activation. Moreover, a constitutively active mutant of PI 3-kinase mimicked insulin's effect ; overexpression of this mutant lead to the inhibition of PEPCK gene transcription in the absence of insulin. These data indicated that PI 3-kinase is necessary and sufficient for insulin's inhibitory effect on PEPCK gene transcription, and the signaling pathways initiated by RAS is not involved in this event.
