Many patients after traumatic brain injury (TBI) complain lack of concentration and memory deficit. To investigate posttraumatic memory deficit, experimentally head-injured rats were examined for the presence of impairment of learning. Fisher-strain male rats of 180 to 260 gm were used. Under anesthesia with ether, the rat's head was fixed on sponge of 3 cm depth. To the head, 40 gm of aluminum bar of 0.9 cm diameter was dropped from 50 cm high level, the bar was guided by plastic tube of 1.0 cm diameter. If we placed a small helmet on the rat's head, fatal contusion or laceration of the skull and of the brain were avoided. Almost all animals injured with helmet showed no prolonged unconsciousness. So head injury, as we modified the original method produced by Manaka et al. was considered to be an experimental model of concussion. After full recovery from the insult, the animals were placed in Iso's computer assisted running wheel apparatus for active avoidance learning. If they stopped running in the wheel cage for 15 sec or more, the computer produced punishment current of 1 mA of 200 vt on the floor grid of the cage. When the number of electric shock (ES) reduced to 0 to 1, we evaluate that the animal learned active avoidance. When the rats have learned this trick, they turn the wheel minimally and avoid ES successfully. As a result, the number of ES was 5.6±2.8 (Mean±SE, n=5) in the head-injured group and 1.7±0.8 (n=6) in the control group. The difference was significant (F = 9.173, P<0.0001 by Two-way repeated-Measures ANOVA). In conclusion, ability of avoidance learning was significantly impaired in the head-injured animals. This experimental result will be applied on further studies in rehabilitation area on the scientific evaluation or training for the individuals with memory deficit in many clinical occasions.
